Clarifying directional dependence among measures of early auditory processing and cognition in schizophrenia: leveraging Gaussian graphical models and Bayesian networks

Background
Research using latent variable models demonstrates that pre-attentive measures of early auditory processing (EAP) and cognition may initiate a cascading effect on daily functioning in schizophrenia. However, such models fail to account for relationships among individual measures of cognition and EAP, thereby limiting their utility. Hence, EAP and cognition may function as complementary and interacting measures of brain function rather than independent stages of information processing. Here, we apply a data-driven approach to identifying directional relationships among neurophysiologic and cognitive variables.
MethodsUsing data from the Consortium on the Genetics of Schizophrenia 2, we estimated Gaussian Graphical Models and Bayesian networks to examine undirected and directed connections between measures of EAP, including mismatch negativity and P3a, and cognition in 663 outpatients with schizophrenia and 630 control participants.
ResultsChain structures emerged among EAP and attention/vigilance measures in schizophrenia and control groups. Concerning differences between the groups, object memory was an influential variable in schizophrenia upon which other cognitive domains depended, and working memory was an influential variable in controls.
ConclusionsMeasures of EAP and attention/vigilance are conditionally independent of other cognitive domains that were used in this study. Findings also revealed additional causal assumptions among measures of cognition that could help guide statistical control and ultimately help identify early-stage targets or surrogate endpoints in schizophrenia.
The relationship between sex and functional outcome in first-episode schizophrenia: the role of premorbid adjustment and insight

Background
Studies that examined sex differences in first-episode patients consistently show that males compared to females have poor premorbid adjustment, earlier age of onset, worse clinical characteristics, and poorer outcomes. However, little is known about potential mediators that could explain these sex differences.
MethodsOur sample consisted of 137 individuals with first episode schizophrenia (males, n = 105; 77%) with a mean age of 22.1(s.d. = 4.1) years and mean education of 12.5(s.d. = 1.7) years. At entry, patients were within 2 years of their first psychotic episode onset. Baseline assessments were conducted for premorbid adjustment, symptoms, cognitive functioning, insight, and at 6-months for role and social functioning.
ResultsMales as compared to females had poorer premorbid adjustment across several key developmental periods (p < 0.01), an earlier age of onset [M = 20.3(3.3) v. 22.8(5.6), p = 0.002], more negative symptoms (p = 0.044), poorer insight (p = 0.031), and poorer baseline and 6-month role (p = 0.002) and social functioning (p = 0.034). Several of these variables in which males showed impairment were significant predictors of 6-month role and social functioning. Premorbid adjustment and insight mediated the relationship between sex and role and social functioning at 6-months, but not negative symptoms.
DiscussionMales compared to females were at lower levels across several key premorbid and clinical domains which are strongly associated with functional outcome supporting the hypothesis that males might have a more disabling form of schizophrenia. The relationship between sex with role and social functioning was mediated through premorbid adjustment and insight suggesting pathways for understanding why females might have a less disabling form of schizophrenia.

Non-technical summary
We summarize some of the past year's most important findings within climate change-related research. New research has improved our understanding about the remaining options to achieve the Paris Agreement goals, through overcoming political barriers to carbon pricing, taking into account non-CO2 factors, a well-designed implementation of demand-side and nature-based solutions, resilience building of ecosystems and the recognition that climate change mitigation costs can be justified by benefits to the health of humans and nature alone. We consider new insights about what to expect if we fail to include a new dimension of fire extremes and the prospect of cascading climate tipping elements.
Technical summaryA synthesis is made of 10 topics within climate research, where there have been significant advances since January 2020. The insights are based on input from an international open call with broad disciplinary scope. Findings include: (1) the options to still keep global warming below 1.5 °C; (2) the impact of non-CO2 factors in global warming; (3) a new dimension of fire extremes forced by climate change; (4) the increasing pressure on interconnected climate tipping elements; (5) the dimensions of climate justice; (6) political challenges impeding the effectiveness of carbon pricing; (7) demand-side solutions as vehicles of climate mitigation; (8) the potentials and caveats of nature-based solutions; (9) how building resilience of marine ecosystems is possible; and (10) that the costs of climate change mitigation policies can be more than justified by the benefits to the health of humans and nature.

Trans-10, cis-12-conjugated linoleic acid (CLA) is a potent bioactive fatty acids (FA) that causes milk fat depression in lactating animals. FA are transferred to milk directly through chylomicrons and indirectly by recycling through other tissues. The objective of this study was to characterise the kinetics of trans-10, cis-12 CLA transfer to plasma and milk after a single bolus infusion. Five multiparous mid-lactation cows received a single abomasal bolus infusion of an enriched CLA mixture providing 15 g of trans-10, cis-12 CLA and 15 g of cis-9, trans-11 CLA over a 30-min period. Plasma concentration of trans-10, cis-12 and cis-9, trans-11 CLA peaked 2 h post-bolus, reaching 0·29 and 0·38 % of total plasma FA, respectively, and returned to pre-bolus values at 72 h post-infusion. Milk trans-10, cis-12 CLA yield and concentration peaked 14 h post-bolus (0·25 g/h) and was not detectable in milk after 86 h. Total apparent transfer of trans-10, cis-12 CLA to milk was 41 %, with 73 % transferred to milk through the direct pool (chylomicrons) and the remaining 27 % transferred through the indirect pool (tissue recycling). Compartmental modelling revealed the existence of a transient unavailable pool of trans-10, cis-12 CLA in extravascular tissues represented primarily by the mammary gland, which slowly exchanges with an available pool for secretion in milk fat and transfer to milk. In conclusion, trans-10, cis-12 CLA is predominantly transferred to milk through the direct pathway; however, how this CLA isomer is processed within the mammary gland requires further investigation.

BACKGROUND
Influenza A (H1N1) pdm09 became the predominant circulating strain in the United States during the 2013–2014 influenza season. Little is known about the epidemiology of severe influenza during this season.
METHODSA retrospective cohort study of severely ill patients with influenza infection in intensive care units in 33 US hospitals from September 1, 2013, through April 1, 2014, was conducted to determine risk factors for mortality present on intensive care unit admission and to describe patient characteristics, spectrum of disease, management, and outcomes.
RESULTSA total of 444 adults and 63 children were admitted to an intensive care unit in a study hospital; 93 adults (20.9%) and 4 children (6.3%) died. By logistic regression analysis, the following factors were significantly associated with mortality among adult patients: older age (>65 years, odds ratio, 3.1 [95% CI, 1.4–6.9], P=.006 and 50–64 years, 2.5 [1.3–4.9], P=.007; reference age 18–49 years), male sex (1.9 [1.1–3.3], P=.031), history of malignant tumor with chemotherapy administered within the prior 6 months (12.1 [3.9–37.0], P<.001), and a higher Sequential Organ Failure Assessment score (for each increase by 1 in score, 1.3 [1.2–1.4], P<.001).
CONCLUSIONRisk factors for death among US patients with severe influenza during the 2013–2014 season, when influenza A (H1N1) pdm09 was the predominant circulating strain type, shifted in the first postpandemic season in which it predominated toward those of a more typical epidemic influenza season.

Cognitive deficits in schizophrenia are increasingly accepted as core features of this disorder that play a role as vulnerability indicators, as enduring abnormalities during clinical remission, and as critical rate-limiting factors in functional recovery. This article demonstrates the lasting influence of Norman Garmezy through his impact on one graduate student and then through his later collaborative research with colleagues. The promise of core cognitive deficits as vulnerability indicators or endophenotypes was demonstrated in research with children born to a parent with schizophrenia as well as with biological parents and siblings of individuals with schizophrenia. In studies of patients with a recent onset of schizophrenia, cognitive deficits were found to endure across psychotic and clinically remitted periods and to have a strong predictive influence on likelihood of returning successfully to work or school. Converging lines of evidence for the enduring core role of cognitive deficit in schizophrenia have led in recent years to a burgeoning interest in developing new interventions that target cognition as a means of improving functional recovery in this disorder.

Host Galaxies of Obscured Quasars: Infant Starbursts Caught in Action

Supermassive black holes (SMBHs) in the centers of massive galaxies are thought to predominantly grow in brief Eddington-rate quasar phases accompanied by starbursts, but on-going starbursts in luminous quasars are difficult to observe. Buried under the natural coronagraph, obscured quasars offer a unique window for direct, robust host-galaxy spectroscopy otherwise virtually inaccessible for luminous quasars. Our pilot study at z ~ 0.5 (Liu et al. 2009) revealed a substantial contribution from very young stellar populations with ages less than ~ 100 Myr in all of the observed host galaxy spectra. More dramatically, in three out of the nine SDSS quasars observed, we have witnessed strong infant starbursts with ages of ~ 5 Myr, clocked by the telltale Wolf–Rayet emission features.

The sul gene of B/rw (Witkin) and 26 independently isolated sul mutants were examined for their suppression of radiation sensitivity. The suppressed lon gene was demonstrated by transducing it into a K12 proC recipient. The sul gene of B/rw and all but one of the B/r mutants was found to be linked to a gene controlling azide resistance. Transduction data established the order of markers as leu azi mutT sul. One B/r mutant was found not transducible with azide resistance, sul mutations were found to suppress the capsular polysaccharide production of lon strains.
exrB: a malB-linked gene in Escherichia coli B involved in sensitivity to radiation and filament formation

PAM 26, a radiation-sensitive mutant of Escherichia coli strain B, is described. Its properties are attributable to a mutation in a gene, exrB, which is cotransducible with malB. It differs from uvrA (also malB-linked) derivatives of strain B in being sensitive to 1-methyl-3-nitro-1-nitroso-guanidine and γ-radiation, and in being able to reactivate UV-irradiated phage T3. It differs from exrA (also malB-linked) derivatives of strain B in forming filaments during the course of normal growth as well as after irradiation. When exrB was transduced into a K12 (lon+) strain, filaments did not form spontaneously. Three-point transductions established the order of markers as met A malB exrB. Based on an analysis of the frequency of wild-type recombinants in a reciprocal transduction between exrA and exrB strains, it was inferred that they are not isogenic and that the order of markers is malB exrA exrB.

Three genetically distinct ochre suppressors have been identified in a strain of Escherichia coli B/r, all of which suppress a tyrosine auxotrophy and classify as class 2 by phage suppression pattern. One ochre suppressor, which was obtained by conversion from a class 2 amber suppressor, and a second ochre suppressor obtained directly from the non-suppressing parent, were found to have separate map locations, though a peculiar phenotype with regard to a leucine auxotrophy is exhibited by strains carrying either suppressor. We suggest that both suppressors correspond to separate genes for glutamine-inserting tRNA. A Leu+ mutant of a strain carrying one of these suppressors was studied and was found to contain a further nonsense suppressor having amber-suppressing activity at a reduced level. We suggest that this suppressor might result from a mutation in another part of the translational machinery concerned with glutamine insertion. The third ochre suppressor has no effect on the leucine auxotrophy and mapping data suggest that it may be supL, an ochre suppressor probably inserting a different amino acid, from glutamine.

In Escherichia coli the u.v. sensitivity gene recA suppressed u.v.-induced filamentation in a lon u.v. sensitive strain without affecting capsular polysaccharide production. recA appears to prevent the stimulus that leads to filamentation in a lon strain.

The effect of ultraviolet radiation (u.v.) on λ lysogens of exrA strains of Escherichia coli was studied. exrA strains could be lysogenized with, as well as support the vegetative reproduction of, λ. However, though spontaneous induction of λ occurred in exrA(λ) strain at 10% the frequency of exrA+(λ) strain, exrA(λ) strains were not induced by u.v. Because λ was not induced in exrA(λ) strains, lysogens of these strains were no more sensitive to u.v. than were non-lysogens.
The heat-inducible mutant λhcI857 could be induced in exrA strains at elevated temperatures. Furthermore, u.v. irradiation of exrA (λhcI857) strain did not prevent the heat induction of this λ mutant. The exrA mutation appeared to interfere only with the inactivation of λ repressor by u.v.
Among the exrA strains studied was strain Bs1(exr A uvrB). Whereas the λ lysogen of strain Bsl could not be induced by u.v. and was no more sensitive to u.v. than its non-lysogen, the exrA+uvrB(λ) derivative of strain Bsl could be induced by u.v. and was more sensitive to u.v. than its non-lysogen.
